Magnetic Beads DNA/RNA Extraction/Magnetic Beads PCR Cleanup kit

The Geneaid Presto™ PureXP Magnetic Beads utilizes unique paramagnetic bead technology for high-throughput purification of PCR amplicons. This system uses an optimized buffer to selectively bind DNA fragments of 100 bp or larger to paramagnetic beads. The protocol consists of binding, washing, and elution steps, effectively removing primers, nucleotides, salts, and enzymes from the reaction mixture, resulting in a more purified PCR product. The purified products can be used for downstream applications, such as PCR, sequencing, cloning, genotyping and library construction for NGS.

QUALITY CONTROL

The quality of the Geneaid Presto™ PureXP PCR purification kit  is tested on a lot-to-lot basis according to Geneaid's ISO-certified quality management system. DNA fragments are purified from PCR products and analyzed by electrophoresis.

Figure 1.The 50 bp DNA ladder was used to simulate PCR amplicons of varying fragment sizes (50-1,500bp, 1000 ng, 20 µL). Purification was performed using Geneaid Presto™ PureXP Magbeads and B brand Magbeads, respectively (in triplicates). The purified products were quantified using a Qubit (Thermo) and analyzed by capillary electrophoresis. The results demonstrated that the Presto™ PureXP Magbeads exhibited a higher recovery rate (84.6%) compared to the B brand (78.6%). Capillary electrophoresis also showed consistent results.
Figure 2.Sheared DNA (green line) was purified separately using Geneaid Presto™ PureXP Magbeads and B brand Magbeads. After adjusting the concentrations of the purified products to the same level, capillary electrophoresis analysis was performed. The results show that the two curves representing Geneaid Presto™ PureXP Magbeads (red line) and Brand B Magbeads (blue line) completely overlap, confirming that the different DNA fragment sizes recovery profile of Geneaid Presto™ PureXP Magbeads is identical to that of B Brand Magbeads.

Figure 3.Sheared DNA was recovered using Geneaid Presto™ PureXP Magbeads with different volume ratios to remove various DNA fragment sizes. After adjusting the concentrations of the purified products to the same level, capillary electrophoresis analysis was performed. The results showed that a Magbead:DNA ratio of 1.8x effectively removed fragments smaller than 100 bp, a ratio of 0.9x removed fragments smaller than 200 bp, 0.7x removed fragments smaller than 250 bp, and 0.6x removed fragments smaller than 300 bp. Geneaid Presto™ PureXP Magbeads demonstrated optimal size selection functionality.
